改良法制作兔VX2骨肿瘤模型的实验研究

目的 通过比较改良法与传统法制作兔VX2骨肿瘤模型,明确改良法可明显改善肿瘤

细胞经骨孔外渗情况。方法 取传代VX2肿瘤荷瘤兔,将肿瘤制备成肿瘤组织块,分别植

入22只成年家兔双侧胫骨内制成兔VX2骨肿瘤模型。所有家兔左侧胫骨采用改良法植入

肿瘤组织块,即通过套管植入肿瘤组织并给予骨蜡封堵;而所有右侧胫骨直接经穿刺骨孔

植入肿瘤组织块。模型制作3w后,给予所有家兔患肢行彩色超声检查,明确有无软组织

包块形成;同时处死家兔,取骨孔周围软组织行病理检查,明确有无肿瘤细胞转移。结果 造

模3w后,双侧胫骨成瘤率100%。超声显示左侧组发现软组织包块2例,右侧组6例;

病理检查示左侧组肿瘤细胞经骨孔外渗率(23%,5/22小于右侧组(55%,12/22)。将

两组穿刺骨孔肿瘤细胞外渗率进行比较,差异有统计学意义(χ2=4.69,P<0.05)。结论 改

良法制作骨肿瘤模型可改善肿瘤细胞经骨孔外渗情况,是一种较为理想的造模方式。

Abstract:Objective To investigate the efficacy between the traditional method

and the improved method in establishment of VX2 bone tumor model in rabbit in

order to prove the improved method can improve tumor cells spreading. Methods

After separating the VX2 tumor from cancer bearing VX2 rabbit, the tumor tissue

blocks were prepared. the tissue blocks were embedded into the right tibias 22

rabbits through the puncture hole ,and all the puncture holes were closed by

bone wax. Color Doppler Ultrasound was used to assess the soft enclosed mass

near the puncture hole after 2 weeks. The pathological study also was carried out

for the soft tissue near the holes after 14 days. Results Only 2 cases from the left

group showed the soft mass around the puncture holes after 14 days, but there

were 6 cases around the puncture holes of all the 22 cases of the right group. The

pathological showed there were 5 cases(23%,5/22) that we can find the tumor

cells in the soft tissue around the puncture hole in the left group, but 12 cases

(55%,12/22)in the right group. The difference was statistically significant

(χ2=4.69,P<0.05). Conclusion The improved method was more convenient in

operation and less tumor cells spreading in establishment of bone tumor model

compared with the traditional method.

目前由于我国人们生活方式及环境因素的改变,肿瘤特别是骨肿瘤的发病率呈逐年升

高趋势。VX2肿瘤一种可移植的恶性乳头状瘤,可移植在兔的肝脏,肺脏,骨骼肌肌肉等

处,所建立的肿瘤模型具有较高的生物学特性[1~3]。由于VX2肿瘤对兔骨质的侵犯及破

坏作用与人类骨肿瘤较为类似,兔VX2骨肿瘤的研究备受学者青睐。良好的兔骨肿瘤模型

是实验研究成功的第一步,然而造模过程中经常遇到肿瘤细胞经骨孔外渗等困难。本实验

中,通过改良法与传统法植入肿瘤细胞制造兔骨肿瘤模型对比,明确改良法可改善肿瘤细

胞经骨孔外渗情况。

1 资料与方法

1.1实验动物 新西兰成年家兔22只,月龄4个月,体质量2~3kg,雌雄不限,所有

动物进行标准兔舍圈养,卫生水及标准兔饲料喂养。本实验遵循承德医学院实验动物保护

及使用规定,遵循医学伦理学要求。

1.2实验器材及仪器 超声诊断扫描仪采用Medical Systems-LOGIQ9(GE公司,美

国);OLYMPUS-BX53光学显微镜(OLYMPUS公司,日本);VX2肿瘤株由广州吉妮欧

生物试剂公司提供。1.3兔VX2肌肉肿瘤模型制备
